چکیده

In multiple hypotheses testing, it is important to control the probability of rejecting “true” null hypotheses. A standard procedure has been to control the family-wise error rate (FWER), the probability of rejecting at least one true null hypothesis. For large numbers of hypotheses, using FWER can result in very low power for testing single hypotheses. Recently, powerful multiple step FDR procedures have been proposed which control the “False Discovery Rate” (expected proportion of Type I errors). More recently van der Laan, Dudoit and Pollard (2004) proposed controlling a generalized family-wise error rate k-FWER (also called gFWER(k)), defined as the probability of at least (k+1) Type I errors (k=0 for the usual FWER). Lehmann and Romano (2005) suggested both a single-step, and a step-down procedure for controlling the generalized family-wise error rate. They make no assumptions concerning the p-values of the individual tests. The step-down procedure is simple to apply, and cannot be improved without violation of control of the k-FWER. In this paper, by limiting the number of steps in step-down or step-up procedures, new procedures are developed to control k-FWER (and the proportion of false positives). Using data from the literature, the procedures are compared with those of Romano and Lehmann (2005), and, under the assumption of a multivariate normal distribution of the test statistics, show considerable improvement in the reduction of the number and proportion of false positives.

A New Proof of FDR Control Based on Forward Filtration

For multiple testing problems, Benjamini and Hochberg (1995) proposed the false discovery rate (FDR) as an alternative to the family-wise error rate (FWER). Since then, researchers have provided many proofs to control the FDR under different assumptions. Storey et al. (2004) showed that the rejection threshold of a BH step-up procedure is a stopping time with respect to the reverse filtration g...

Multiple testing. Part I. Single-step procedures for control of general type I error rates.

The present article proposes general single-step multiple testing procedures for controlling Type I error rates defined as arbitrary parameters of the distribution of the number of Type I errors, such as the generalized family-wise error rate. A key feature of our approach is the test statistics null distribution (rather than data generating null distribution) used to derive cut-offs (i.e., rej...

An Adaptive Step - down Procedure with Proven Fdr Control under Independence

In this work we study an adaptive step-down procedure for testing m hypotheses. It stems from the repeated use of the false discovery rate controlling the linear step-up procedure (sometimes called BH), and makes use of the critical constants iq/[(m + 1− i(1− q)], i= 1, . . . ,m. Motivated by its success as a model selection procedure, as well as by its asymptotic optimality, we are interested ...

FDR Step-down and Step-up Procedures for the Correlated Case

Controlling the false discovery rate has been increasingly utilized in high dimensional screening studies where multiplicity is a problem. Most methods do not explicitly take the correlation between the data or the test statistics into account, with consequent loss of power. In this paper, we use least favorable configurations to obtain critical values for both step-down and step-up procedures,...
